Here's the key thing to understand about this pairing: it's the melatonin that gets affected, not your birth control pill. Enovid-E (a combination of mestranol, an estrogen, and norethynodrel, a progestin) can raise your body's own melatonin levels and slow how fast melatonin is cleared. So when you add a melatonin supplement on top of that, the melatonin can stack up and hit you harder than expected.

In real life that usually means more of melatonin's typical effects: extra drowsiness, grogginess the next morning, vivid dreams, or feeling a bit dizzy. It does not make your contraceptive less effective or change how it protects you. This is mostly theoretical and based on limited evidence, so don't be alarmed. If you want to use melatonin, just check with your pharmacist first, and consider starting low.

Theoretically, melatonin might increase levels of drugs metabolized by CYP3A4.
Laboratory research shows that certain lots of melatonin inhibit CYP3A4. Theoretically, combined administration of melatonin with certain drugs metabolized by CYP3A4 may reduce the metabolism of these drugs, resulting in increased serum levels. Until more is known, use melatonin cautiously in patients taking drugs metabolized by these enzymes.

Enovid-E is an older birth control pill that combines an estrogen (mestranol) with a progestin (norethynodrel). Both hormones are processed in part by a liver enzyme called CYP3A4. The idea here is that melatonin might slow that enzyme down a little, which could let these hormones build up slightly and add to everyday side effects like nausea, breast tenderness, headache, or bloating.

Here's the reassuring part: this concern comes only from lab testing on certain batches of melatonin, not from real people, and it's rated as a minor, theoretical interaction. For most folks, short-term melatonin for sleep is unlikely to cause a noticeable problem. Still, since hormone levels matter, it's worth a quick mention to your pharmacist or doctor before pairing them.
