Amitriptyline, Perphenazine falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES — the group N,n-dmpea interacts with.

In vitro research shows that N,N-DMPEA strongly inhibits cytochrome P450 2D6 (CYP2D6). Theoretically, concomitant use with drugs metabolized by CYP2D6 might increase the risk for adverse effects from these drugs.
Some drugs metabolized by CYP2D6 include amitriptyline (Elavil), clozapine (Clozaril), codeine, desipramine (Norpramin), donepezil (Aricept), fentanyl (Duragesic), flecainide (Tambocor), fluoxetine (Prozac), meperidine (Demerol), methadone (Dolophine), metoprolol (Lopressor, Toprol XL), olanzapine (Zyprexa), ondansetron (Zofran), tramadol (Ultram), trazodone (Desyrel), and others.

This combo pill pairs amitriptyline (a tricyclic antidepressant) with perphenazine (an antipsychotic). Both lean heavily on a liver enzyme called CYP2D6 to be cleared from your body. In a test-tube setting, N,N-DMPEA appears to strongly block that enzyme. If that holds true in people, the medication could clear more slowly and build up, pushing up the chance of side effects like heavy drowsiness, dry mouth, constipation, blurred vision, dizziness when standing, a faster or irregular heartbeat, and muscle stiffness or restlessness from the perphenazine side.

Keep in mind this is theory based on lab work, not human studies, and N,N-DMPEA itself is poorly studied with its own stimulant effects. Please don't change anything on your own. Run it past your pharmacist or doctor first.
