Why this applies to Perphenazine

Perphenazine falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES — the group N,n-dmpea interacts with.

In vitro research shows that N,N-DMPEA strongly inhibits cytochrome P450 2D6 (CYP2D6). Theoretically, concomitant use with drugs metabolized by CYP2D6 might increase the risk for adverse effects from these drugs.
Some drugs metabolized by CYP2D6 include amitriptyline (Elavil), clozapine (Clozaril), codeine, desipramine (Norpramin), donepezil (Aricept), fentanyl (Duragesic), flecainide (Tambocor), fluoxetine (Prozac), meperidine (Demerol), methadone (Dolophine), metoprolol (Lopressor, Toprol XL), olanzapine (Zyprexa), ondansetron (Zofran), tramadol (Ultram), trazodone (Desyrel), and others.

Here's the gist for perphenazine. Your liver uses an enzyme called CYP2D6 to help clear this medication. Lab testing suggests N,N-DMPEA can block that enzyme. If it does the same thing in people, perphenazine could clear more slowly and build up in your body, which may mean stronger effects and a higher chance of side effects like drowsiness, dizziness, low blood pressure, a faster heartbeat, or those uncomfortable muscle and movement reactions (stiffness, restlessness, tremor) that older antipsychotics can cause.

I want to be honest that this is theoretical, based only on test-tube research, not real human studies. N,N-DMPEA itself is poorly studied. Please don't change anything on your own. Run this combo by your pharmacist or doctor first.

Perphenazine tablets are approved to treat schizophrenia in adults. They are also used to control severe nausea and vomiting in adults when other measures are not enough.

Perphenazine has not been shown effective for managing behavioral problems in patients with intellectual disabilities, and it is not approved for dementia-related psychosis in elderly patients.
