Here's the gist: Limbitrol DS combines amitriptyline (an older antidepressant) with chlordiazepoxide (a benzodiazepine for anxiety). Neither is a prodrug, so they work as-is once you take them. The concern with adrafinil is that your liver turns it into modafinil, and modafinil can gently slow down a liver enzyme called CYP2C19, which helps clear part of this medication. If that clearing slows, the drug could build up a bit and lead to more pronounced effects like daytime drowsiness, grogginess, dry mouth, constipation, dizziness, or a faster heartbeat.

That said, this is a theoretical interaction based mostly on lab work, not real-world reports, so I wouldn't lose sleep over it. Still, please check with your pharmacist or doctor before pairing them.

Adrafinil is a precursor to modafinil, and modafinil may slightly speed up a liver enzyme called CYP1A2. Amitriptyline is partly handled by that enzyme, so in theory, nudging it faster could lower amitriptyline levels and make it work a little less well for mood or nerve pain.

I'll be honest though: this is theoretical, based on lab data showing only weak enzyme activity, so it may not amount to much in real life. The bigger practical concern is that modafinil is a stimulant while chlordiazepoxide is a sedative, which can pull against each other. Please run this combo by your doctor or pharmacist first.
