Clopidogrel falls under CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES — the group Adrafinil interacts with.
Adrafinil is a precursor to the stimulant prescription drug modafinil. Modafinil has been shown to weakly induce CYP1A2 activity in vitro.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's something worth knowing about pairing these two. Adrafinil turns into modafinil in your body, and modafinil may mildly speed up a liver enzyme called CYP1A2. The concern on file is that this could lower the levels of drugs broken down by that enzyme. The good news for Plavix is that clopidogrel doesn't really rely on CYP1A2 to do its job. Clopidogrel is a prodrug activated mainly through CYP2C19, so this particular pathway isn't the one that matters most for it.
This is a theoretical note based on lab data, not real-world reports, so I wouldn't lose sleep over it. Still, since clopidogrel protects you from clots, please run any new supplement past your pharmacist or doctor first.

Adrafinil is a precursor to the stimulant prescription drug modafinil. Modafinil has been shown to reduce CYP2C9 expression in vitro.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the wrinkle with Plavix (clopidogrel): it's a prodrug, meaning it does nothing until your liver enzymes flip it into its active, clot-fighting form. So the usual "this supplement raises drug levels" logic gets turned on its head. Adrafinil becomes modafinil in your body, and modafinil may slow down CYP2C9, one of the enzymes that helps activate clopidogrel. If less drug gets activated, you could end up with weaker blood-thinning protection, which matters a lot when the whole point is preventing another stroke, heart attack, or stent clot.
I'll be honest, this is theoretical and based only on lab data, not real patient reports, so don't panic. Still, because clopidogrel is protecting against serious events, please run any adrafinil use by your pharmacist or doctor first.

Here's a wrinkle worth knowing: clopidogrel (Plavix) is a prodrug, meaning your liver has to switch it on before it can thin your blood and protect against clots. While the enzyme CYP2C19 does most of that activation, CYP3A4 also plays a part. Adrafinil turns into modafinil in your body, and modafinil can mildly speed up (induce) CYP3A4.
In theory, that faster enzyme could change how much active clopidogrel you make. The honest answer is that the direction is uncertain and this is only a theoretical concern based on how these drugs work, not real-world reports. Since Plavix is keeping your blood from clotting after a stroke, heart attack, or stent, even a small shift matters. Please run this by your doctor or pharmacist before combining them.

What Is Clopidogrel Used For?

Clopidogrel tablets are used to reduce the risk of heart attack and stroke in people with acute coronary syndrome (ACS) — including unstable angina and non-ST-elevation or ST-elevation heart attacks — and in people with a history of recent heart attack, recent stroke, or established peripheral arterial disease. For ACS, it is typically given alongside aspirin.
It works by preventing platelets (blood cells that trigger clotting) from clumping together, which helps keep arteries open and lowers the chance of a dangerous clot forming.
