Esomeprazole Strontium falls under CYTOCHROME P450 2C19 (CYP2C19) SUBSTRATES — the group Adrafinil interacts with.

Theoretically, adrafinil might increase the levels and clinical effects of CYP2C19 substrates.
Adrafinil is a precursor to the stimulant prescription drug modafinil. Modafinil has been shown to inhibit CYP2C19 activity in vitro.

Here's the gist of this pairing. Your body turns adrafinil into modafinil, and modafinil can gently slow down a liver enzyme called CYP2C19. Esomeprazole (the acid-reducer in Esomeprazole Strontium) is cleared partly by that same enzyme, so in theory the modafinil could let esomeprazole build up a bit higher than usual.

If that happened, you might notice slightly stronger acid-blocking effects or mild side effects like headache, nausea, or stomach upset. Honestly though, this is a theoretical concern based on lab testing, not real patient reports, and esomeprazole is a pretty forgiving drug, so it's unlikely to cause trouble. Still, since adrafinil is unregulated and hard on the liver, please run this combo by your pharmacist or doctor before mixing them.

Here's the gist for this pairing. Once your liver turns adrafinil into modafinil, that modafinil can mildly speed up an enzyme called CYP3A4. Since esomeprazole strontium (a heartburn medicine that lowers stomach acid) is partly broken down by that same enzyme, faster clearance could leave a little less of it in your system. In plain terms, the worry is that your acid reflux protection might work a bit weaker, so you could notice symptoms like heartburn or stomach burning creeping back.

I'd keep this in perspective, though. This is a theoretical interaction based on how the drugs behave, not on real patient reports, and the effect is described as weak. Still, since adrafinil is unregulated and hard on the liver, please run it by your pharmacist or doctor before combining the two.
