Riluzole falls under CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES — the group Adrafinil interacts with.

Theoretically, adrafinil might reduce the levels and clinical effects of CYP1A2 substrates.
Adrafinil is a precursor to the stimulant prescription drug modafinil. Modafinil has been shown to weakly induce CYP1A2 activity in vitro.

Here's the situation with these two. Your body turns adrafinil into modafinil, and modafinil may slightly speed up a liver enzyme called CYP1A2. Riluzole leans heavily on that same enzyme to be cleared from your body. So in theory, taking adrafinil could nudge your liver to break down riluzole a bit faster, leaving less in your system to do its job protecting your nerves in ALS.

I want to be honest about how soft this is. This is a theoretical concern from lab data only, and the effect on the enzyme is described as weak. We don't have real human cases showing riluzole stopped working. Still, because riluzole matters so much in ALS, please loop in your doctor or pharmacist before pairing these.

What Is Riluzole Used For?

Riluzole is approved to treat amyotrophic lateral sclerosis (ALS) — the progressive neurological disease also known as Lou Gehrig's disease. This indication is shared across all available riluzole products, including tablets (Rilutek, riluzole), the dissolving oral film (Exservan), and the oral suspension (Tiglutik).

Tiglutik oral suspension is notable in that it can also be given through a PEG-tube (percutaneous endoscopic gastrostomy tube) for patients who have difficulty swallowing.
