Theoretically, adrafinil might increase the levels and clinical effects of CYP2C9 substrates.
Adrafinil is a precursor to the stimulant prescription drug modafinil. Modafinil has been shown to reduce CYP2C9 expression in vitro.

Here's the gist for these two. Your liver uses an enzyme called CYP2C9 to help clear sildenafil (Viagra, Revatio) out of your system. Adrafinil turns into modafinil in the body, and lab studies hint that modafinil may slow this particular enzyme down a bit. If that happens, sildenafil could hang around longer and feel stronger than usual.

In real life that might mean more of sildenafil's typical effects, like headache, flushing, a stuffy nose, dizziness, or a bigger drop in blood pressure. I want to be honest though: this is theoretical, based only on test-tube data, so it may not amount to much in people. Still, since adrafinil itself is unregulated and hard on the liver, please run this combo by your doctor or pharmacist first.

Here's the gist for sildenafil (Viagra or Revatio). Your body clears this drug using a liver enzyme called CYP3A4. Once adrafinil turns into modafinil in your liver, that modafinil can gently speed up that same enzyme. The likely result is that sildenafil gets broken down a bit faster, so there may be less of it working in your system.

In plain terms, that could mean a weaker effect: less help with an erection, or less benefit for pulmonary hypertension if that's why you take it. Sildenafil is not a prodrug, so this points toward reduced effect, not extra side effects. Honestly, this is a theoretical interaction based on how the drugs behave, and modafinil is only a weak enzyme booster, so it may not amount to much. Still, run it by your pharmacist or doctor before combining them.

What Is Sildenafil Used For?

Brand name: Revatio

Sildenafil treats two distinct conditions depending on the product. Viagra (tablet) and Vybrique (dissolving oral film) are approved for erectile dysfunction (ED) — difficulty getting or keeping an erection. Generic sildenafil tablets and oral film are also approved for ED.

Revatio and several generic sildenafil products (tablets, oral suspension, and injection) are used for pulmonary arterial hypertension (PAH) — high blood pressure in the arteries of the lungs — in adults to improve exercise ability and slow the disease from getting worse. Revatio oral suspension and injection are also approved for PAH in children ages 1 to 17.
