Caffeine and Rabeprazole

Here is the interaction we found between these two, with the evidence behind it.

Minor

CYTOCHROME P450 1A2 (CYP1A2) INHIBITORS

Why this applies to Rabeprazole

Rabeprazole falls under CYTOCHROME P450 1A2 (CYP1A2) INHIBITORS — the group Caffeine interacts with.

Theoretically, concomitant use might increase the levels and adverse effects of caffeine.
Caffeine is metabolized by CYP1A2. Theoretically, drugs that inhibit CYP1A2 may decrease the rate of caffeine clearance and increase caffeine levels.
Severity Minor Likelihood Possible Evidence D

Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.

Dr. Brian Staiger, PharmD, BCPS
Dr. Brian Staiger explains PharmD, Clinical Pharmacist

Here's the thing to keep in mind with this pairing: it's really about the caffeine, not the rabeprazole. Your liver uses an enzyme called CYP1A2 to clear caffeine out of your system. Rabeprazole can mildly slow that enzyme down, so in theory caffeine could hang around a little longer and feel a bit stronger. That might mean more jitteriness, a racing heart, or trouble sleeping if you've had a lot of coffee, tea, or energy drinks.

I want to be honest, though, this is only a theoretical concern based on lab studies, not real patient cases, and it's rated as minor. Your rabeprazole will still work fine for your heartburn or reflux. If you notice caffeine hitting you harder than usual, just ease up on it and mention it to your pharmacist or doctor.

  • LikelyWell-controlled human studies have demonstrated the likely existence of this interaction.
  • ProbableInteraction has not been documented in well-controlled studies, however, the interaction has been demonstrated in some small human studies or in controlled animal studies in conjunction with multiple case reports.
  • PossibleInteraction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
  • UnlikelyInteraction has been demonstrated in animal or in lab research but has been shown not to occur in humans.
Evidence the quality of the studies behind the rating (graded A–D)
  • AHigh-quality meta-analysis (quantitative systematic review).

From our pharmacist’s overview on the Rabeprazole page

“Rabeprazole (Pariet) quiets stomach acid for reflux and ulcers, but its supplement list deserves genuine care, with three serious entries to understand before anything else. Most items matter because they change how the liver breaks rabeprazole down, so its level can drift up or down.”

About Rabeprazole

What Is Rabeprazole Used For?

Brand name: Pariet

Rabeprazole delayed-release tablets treat a range of acid-related conditions in adults, including healing and maintenance of erosive or ulcerative GERD (gastroesophageal reflux disease), symptomatic GERD, duodenal ulcers, and pathological hypersecretory conditions such as Zollinger-Ellison syndrome.
