Ethinyl Estradiol, Norethindrone falls under CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES — the group Melatonin interacts with.
Melatonin is metabolized in the liver primarily by the CYP2C19 and CYP1A2 enzymes. Theoretically, combined administration of melatonin with drugs metabolized by the CYP1A2 enzyme might reduce the metabolism of these drugs, resulting in increased serum levels. Conversely, some drugs metabolized by CYP1A2 may inhibit the metabolism of melatonin, resulting in increased serum levels of melatonin. Until more is known, use melatonin cautiously in patients taking drugs metabolized by these enzymes.
Interaction has not been documented in well-controlled studies, however, the interaction has been demonstrated in some small human studies or in controlled animal studies in conjunction with multiple case reports..
Here's the gist for your birth control pill (the ethinyl estradiol and norethindrone combo). The estrogen part is partly broken down by a liver enzyme called CYP1A2. On paper, melatonin might slow that enzyme down a touch, which could let a little more estrogen linger in your system. If anything happened, you'd be looking at mild estrogen-type effects like nausea, breast tenderness, bloating, or some breakthrough spotting.
The good news: this is purely theoretical, based on how the drugs behave in the lab, not on real reports of problems in people. Your pill is not a prodrug, so it still works normally for preventing pregnancy. Most people use both with no trouble, but it never hurts to mention it to your pharmacist or doctor.

Here's the thing to know about combining your birth control pill (the ethinyl estradiol and norethindrone in products like Loestrin or Ovcon) with a melatonin supplement: it's really the melatonin side we watch, not your contraceptive. The estrogen in your pill can slow how fast your body breaks down melatonin, so the melatonin can hang around a bit longer and feel stronger than usual.
In plain terms, you might feel more drowsy, groggy in the morning, or a little lightheaded. This is considered fairly likely but on the milder side, and the evidence behind it is only moderate. A practical tip is to start with a low dose and see how you feel. Your birth control should still work fine. If the grogginess bothers you, check with your pharmacist or doctor.

Laboratory research shows that certain lots of melatonin inhibit CYP3A4. Theoretically, combined administration of melatonin with certain drugs metabolized by CYP3A4 may reduce the metabolism of these drugs, resulting in increased serum levels. Until more is known, use melatonin cautiously in patients taking drugs metabolized by these enzymes.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
