Ethinyl Estradiol, Norgestimate falls under CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES — the group Melatonin interacts with.

Theoretically, melatonin might increase levels of drugs metabolized by CYP1A2. Also, other CYP1A2 substrates might decrease the metabolism of melatonin, increasing melatonin levels.
Melatonin is metabolized in the liver primarily by the CYP2C19 and CYP1A2 enzymes. Theoretically, combined administration of melatonin with drugs metabolized by the CYP1A2 enzyme might reduce the metabolism of these drugs, resulting in increased serum levels. Conversely, some drugs metabolized by CYP1A2 may inhibit the metabolism of melatonin, resulting in increased serum levels of melatonin. Until more is known, use melatonin cautiously in patients taking drugs metabolized by these enzymes.

Here's the gist for your birth control pill (Ortho Tri-Cyclen and similar combos of ethinyl estradiol and norgestimate). On paper, melatonin might slow down one of the liver enzymes (CYP1A2) that helps clear the estrogen part of your pill. If that happens, estrogen levels could nudge a little higher, which in theory might mean more of the usual estrogen-related side effects like nausea, breast tenderness, bloating, or headaches.

I want to be honest, though: this is a theoretical concern based on how the drugs are processed, not something proven in real patients. For most people it's unlikely to cause trouble, and there's no clear sign it weakens your birth control. Still, it's worth a quick chat with your pharmacist or doctor before pairing them regularly.

Here's the key thing to know: this interaction works in one direction. Your birth control pill (Ortho Tri-Cyclen and similar brands) tends to slow down how your body clears melatonin, so taking a melatonin supplement on top of the pill can leave more melatonin in your system than usual. In plain terms, the melatonin may hit a little harder than expected.

What that usually looks like is stronger melatonin-type effects: more daytime grogginess, drowsiness, a bit of a hangover feeling in the morning, or maybe a mild headache. The pill itself isn't being weakened, and your contraception protection isn't the concern here. If you take both, a lower melatonin dose to start often makes sense. Check with your pharmacist or doctor before adjusting anything.

Here's the short version for your birth control pill (Ortho Tri-Cyclen and similar brands): your body uses an enzyme called CYP3A4 to help break down the estrogen part, ethinyl estradiol. Some lab studies hint that certain batches of melatonin might slow that enzyme down a little. In theory, that could let estrogen levels creep up slightly, which might mean a touch more of the usual estrogen side effects like nausea, breast tenderness, or headache.

I want to be honest with you, though: this is very weak evidence, based only on test-tube research, and it's rated as a minor interaction.
