Melatonin + Lioresal
Lioresal brand name: Lioresal Injection.
Melatonin has a moderate interaction with Lioresal. Theoretically, taking melatonin with drugs that lower the seizure threshold might increase the risk of seizure activity.

Here is the interaction we found between these two, with the evidence behind it.
SEIZURE THRESHOLD LOWERING DRUGS
Why this applies to Lioresal
Lioresal falls under SEIZURE THRESHOLD LOWERING DRUGS — the group Melatonin interacts with.
Some clinical evidence suggests that melatonin may increase the frequency of seizures in certain patients, particularly children with neurological disabilities.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the deal with combining melatonin and Lioresal (baclofen). Baclofen isn't a sedative in the usual sense, but it works on your spinal cord nerves and can lower the body's seizure threshold a bit, meaning it can make seizures slightly easier to trigger in people who are prone to them. Melatonin, in theory, may also nudge seizure risk upward in certain people. Put them together and that small effect could add up.
I want to be honest about the strength of this one: it's largely theoretical, based on how these compounds behave rather than strong clinical proof. The signal is clearest in children with neurological conditions. For most adults using occasional melatonin, the risk is likely low, but if you have a seizure history, please run it by your pharmacist or doctor first.

From our pharmacist’s overview on the Lioresal page
“Lioresal is baclofen, a muscle relaxant prescribed for spasticity, and I would treat its supplement list with genuine care. Of the ten ingredients on file, six, including everyday picks like melatonin and ginkgo, may lower the seizure threshold, an added worry with a drug whose abrupt changes can already provoke seizures.”

About Melatonin
What is Melatonin?
- Trouble falling asleep (insomnia)
- Jet lag
- Delayed sleep phase syndrome
- Non-24-hour sleep-wake disorder
- Shift-work sleep problems
- Sleep problems in some children with conditions like autism or ADHD
Melatonin is a hormone your body makes naturally to help control your sleep-wake cycle, and the supplement form is widely used to help with sleep timing problems and jet lag. The evidence is strongest for certain circadian (body-clock) disorders and is more mixed for everyday insomnia. It is generally well tolerated for short-term use, but talk with your pharmacist or doctor before starting, especially for children or during pregnancy.
About Baclofen
What Is Baclofen Used For?
Brand name: Lioresal Injection
Baclofen treats spasticity, which means stiff, tight muscles with spasms. Oral baclofen (tablets, Baclofen oral solution, Ozobax, Baclofen oral suspension, Fleqsuvy, Lyvispah) is used for spasticity from multiple sclerosis, especially flexor spasms with pain, clonus (rhythmic jerking) and muscle rigidity. It may also help some people with spinal cord injuries and other spinal cord diseases.
Intrathecal baclofen (Gablofen, Lioresal Intrathecal and other baclofen injections) is used for severe spasticity. It is given into the fluid around the spinal cord, usually through an implanted pump. It is not for skeletal muscle spasm from rheumatic disorders, and baclofen tablets are not recommended for stroke, cerebral palsy or Parkinson’s disease.
