Here's the connection: Pediazole contains erythromycin, and erythromycin is known to slow down a liver enzyme called CYP3A4. That same enzyme is responsible for clearing modafinil out of your body, and modafinil is what your liver turns adrafinil into. So in theory, taking these together could let modafinil build up a bit more than usual, which might mean stronger stimulant effects like trouble sleeping, jitteriness, a racing heart, headache, or feeling on edge.

I want to be honest with you though: this is a theoretical concern based on how these compounds behave, not on solid studies in people. Adrafinil is also unregulated and can be hard on the liver, so I'd really encourage you to run this combination by your pharmacist or doctor before mixing them.

Here's the gist for Pediazole, which combines erythromycin (an antibiotic) with sulfisoxazole. The erythromycin part is processed by a liver enzyme called CYP3A4. Adrafinil turns into modafinil in your body, and modafinil can gently speed up that same enzyme. In theory, that could clear erythromycin a little faster and make it slightly less effective at fighting your infection.

I want to be honest about how soft this is: it's based on pharmacology reasoning, not real-world reports, and the effect on CYP3A4 is described as weak. Still, when you're treating an infection, you don't want anything nudging the antibiotic down. Since adrafinil is unregulated and hard on the liver, please run this combination by your pharmacist or doctor before mixing them.
