Theoretically, adrafinil might increase the levels and clinical effects of CYP2C19 substrates.
Adrafinil is a precursor to the stimulant prescription drug modafinil. Modafinil has been shown to inhibit CYP2C19 activity in vitro.

Lofepramine is a tricyclic antidepressant, and part of how your body clears it relies on a liver enzyme called CYP2C19. Here's the wrinkle: once adrafinil turns into modafinil in your liver, modafinil can slow that same enzyme down. In theory, that could let lofepramine build up a bit higher than usual.

If levels climb, you might notice more of lofepramine's typical side effects, things like dry mouth, constipation, blurry vision, dizziness, drowsiness, or a faster heartbeat. I want to be honest though: this is a theoretical concern based on lab data, not something proven in real patients, so don't panic. Still, before combining these, please check with your doctor or pharmacist first.

Here's the gist of it. Lofepramine is a tricyclic antidepressant, and part of how your body clears it runs through a liver enzyme called CYP1A2. Adrafinil turns into modafinil in your body, and modafinil may gently speed up that enzyme. The theory is that this could clear your lofepramine a bit faster, leaving slightly less of it around to do its job.

In plain terms, that might mean your antidepressant works a little less well, so a low mood or sleep troubles could creep back. I'll be honest though, this is only a theoretical concern based on lab data, not real-world reports, and the effect looks weak. Still, since adrafinil is unregulated and rough on the liver, please run it by your doctor or pharmacist before combining the two.
