rosiglitazone, glimepiride falls under CYTOCHROME P450 2C9 (CYP2C9) SUBSTRATES — the group Adrafinil interacts with.

Theoretically, adrafinil might increase the levels and clinical effects of CYP2C9 substrates.
Adrafinil is a precursor to the stimulant prescription drug modafinil. Modafinil has been shown to reduce CYP2C9 expression in vitro.

Here's the gist for Avandaryl, which pairs rosiglitazone with glimepiride. Both pieces are partly cleared by a liver enzyme called CYP2C9. Adrafinil turns into modafinil in your body, and in lab studies modafinil seems to slow that enzyme down a bit. If that happens, these medicines could linger longer and hit harder than expected.

The part I'd watch most is the glimepiride, since it actively lowers blood sugar. A stronger-than-usual effect could nudge you toward low blood sugar (shakiness, sweating, dizziness, confusion). Honestly though, this is theoretical, based only on how the drugs behave in test tubes, not real patient reports. Still, please run it by your pharmacist or doctor before mixing them, and keep an eye on your sugar readings.
