Colchicine, Probenecid falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Cascara Sagrada interacts with.

Theoretically, cascara sagrada might decrease the effects of CYP3A4 substrates.
In vitro research suggests that cascara sagrada can induce CYP3A4 enzymes, albeit to a much lower degree than rifampin, a known CYP3A4 inducer.

Here's the situation with your ColBenemid (colchicine plus probenecid) and cascara sagrada. The colchicine part is cleared from your body partly by an enzyme called CYP3A4. In lab dish studies, cascara seemed to gently speed up that enzyme, which in theory could clear colchicine a little faster and make it slightly less effective for gout. I want to stress how weak this is though: it's only test-tube data, the effect was far smaller than known enzyme boosters, and it's rated a minor, possible interaction. Colchicine is not a prodrug, so faster clearance would mean less drug, not more.

Honestly, the bigger real-world concern is the laxative itself. Cascara can cause cramping, dehydration, and shifts in your potassium and other electrolytes, and that matters when you're on these medications. Please check with your pharmacist or doctor before using it.
