Why this applies to Diazepam

Diazepam falls under CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES — the group Ceylon Leadwort interacts with.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 1A2 (CYP1A2) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP1A2. Some substrates of CYP1A2 include clozapine (Clozaril), cyclobenzaprine (Flexeril), fluvoxamine (Luvox), haloperidol (Haldol), imipramine (Tofranil), mexiletine (Mexitil), olanzapine (Zyprexa), pentazocine (Talwin), propranolol (Inderal), tacrine (Cognex), zileuton (Zyflo), zolmitriptan (Zomig), and others.

Here's the gist for Valium (diazepam) with Ceylon Leadwort. The plant compound plumbagin can slow down a liver enzyme called CYP1A2. When that enzyme is blocked, drugs cleared through it can build up a bit higher than usual, which in theory could make a benzodiazepine like diazepam feel stronger. That might mean extra drowsiness, dizziness, foggy thinking, or weak, wobbly muscles.

I'll be honest though, this concern is based only on lab studies, not on real people, so it's more of a theoretical caution than a proven problem. Diazepam also leans more heavily on other liver enzymes (CYP2C19 and CYP3A4) than on CYP1A2. Still, since both can cause sedation, please check with your pharmacist or doctor before combining them, and never stop your diazepam on your own.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 2C9 (CYP2C9) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP2C9. Drugs that are metabolized by CYP2C9 include celecoxib (Celebrex), diclofenac (Voltaren), fluvastatin (Lescol), glipizide (Glucotrol), ibuprofen (Advil, Motrin), irbesartan (Avapro), losartan (Cozaar), phenytoin (Dilantin), piroxicam (Feldene), tamoxifen (Nolvadex), tolbutamide (Tolinase), torsemide (Demadex), and S-warfarin (Coumadin).

Here's the gist: a compound in Ceylon Leadwort called plumbagin can block a liver enzyme (CYP2C9) in lab studies. If that same blocking happened in people, drugs cleared by that enzyme could build up. Diazepam (Valium) is an active medicine, not a prodrug, so higher levels would mean a stronger effect, think extra drowsiness, dizziness, foggy thinking, or trouble with balance.

That said, I want to be honest about the strength of evidence here. This has only been seen in test-tube and lab work, never reported in actual patients, so it's theoretical. On top of that, diazepam leans more on other enzymes (like CYP3A4 and CYP2C19) than on CYP2C9. Still, before pairing them, check with your pharmacist or doctor.

Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP3A4. Some drugs metabolized by CYP3A4 include lovastatin (Mevacor), clarithromycin (Biaxin), indinavir (Crixivan), sildenafil (Viagra), triazolam (Halcion), and numerous others.

Ceylon Leadwort (Plumbago zeylanica) is a plant long used in Ayurvedic and other traditional medicine systems, mainly for digestion, skin problems, and pain. Solid human studies are lacking, and the plant contains plumbagin, a compound that can irritate skin and tissues and may be unsafe in larger amounts. What Is Diazepam Used For?

  • Diazepam is used to relieve anxiety and to control agitation caused by alcohol withdrawal. It is also used along with other medications to control muscle spasms and spasticity caused by certain neurological disorders such as cerebral palsy (condition that causes difficulty with movement and balance), paraplegia (inability to move parts of the body), athetosis (abnormal muscle contractions), and stiff-man syndrome (a rare disorder with muscle rigidity and stiffness). Diazepam is also used along with other medications to control seizures. Diazepam is in a class of medications called benzodiazepines. It works by calming abnormal overactivity in the brain.
