Why this applies to Orphenadrine

Orphenadrine falls under CYTOCHROME P450 2B6 (CYP2B6) SUBSTRATES — the group Ceylon Leadwort interacts with.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 2B6 (CYP2B6) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP2B6. Drugs that are metabolized by CYP2B6 include ketamine (Ketalar), phenobarbital, orphenadrine (Norflex), secobarbital (Seconal), and dexamethasone (Decadron).

Here's the short version on pairing Ceylon Leadwort with your orphenadrine (Norflex). Your liver uses an enzyme called CYP2B6 to help clear orphenadrine out of your system. In test-tube studies, plumbagin (a compound in Ceylon Leadwort) appears to slow that same enzyme down. If that held true in people, orphenadrine could clear more slowly and build up a bit, which might make its anticholinergic side effects more noticeable, things like dry mouth, blurred vision, constipation, trouble urinating, dizziness, or extra drowsiness.

I want to be honest with you, though: this is purely laboratory evidence right now, and it hasn't been seen in actual patients. So it's more of a theoretical caution than a sure thing. Still, before combining the two, run it by your pharmacist or doctor.

Orphenadrine citrate is used to ease discomfort from acute, painful musculoskeletal conditions — things like muscle pain and stiffness. It is always used as part of a broader treatment plan that includes rest and physical therapy, not as a stand-alone cure.

Both the extended-release tablet and the injection form are approved for this use. Neither form directly relaxes tight muscles — the relief it provides is thought to be related to its pain-relieving (analgesic) properties.
