Glyburide falls under CYTOCHROME P450 2C9 (CYP2C9) SUBSTRATES — the group Ceylon Leadwort interacts with.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 2C9 (CYP2C9) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP2C9. Drugs that are metabolized by CYP2C9 include celecoxib (Celebrex), diclofenac (Voltaren), fluvastatin (Lescol), glipizide (Glucotrol), ibuprofen (Advil, Motrin), irbesartan (Avapro), losartan (Cozaar), phenytoin (Dilantin), piroxicam (Feldene), tamoxifen (Nolvadex), tolbutamide (Tolinase), torsemide (Demadex), and S-warfarin (Coumadin).

Here's the gist with Ceylon Leadwort and glyburide. Your body uses an enzyme called CYP2C9 to break down glyburide and clear it out. A compound in this plant, plumbagin, has been shown to slow that enzyme down in lab dishes. If it does the same thing in people, glyburide could hang around longer and hit harder than usually expected.

For a blood sugar medicine, that mostly means a higher chance of low blood sugar, so watch for shakiness, sweating, dizziness, confusion, or a racing heart. The reassuring part: this is only seen in test-tube research, not in real patients, so it's theoretical for now. Still, I'd loop in your pharmacist or doctor before pairing them, and never change your glyburide dose on your own.

Ceylon leadwort root extract has been shown to both increase and decrease blood glucose levels in animal research. This effect has not yet been demonstrated in humans. Theoretically, Ceylon leadwort might reduce or potentiate the effects of antidiabetes drugs. Monitor blood glucose levels closely. Medication dose adjustments may be necessary. Some antidiabetes drugs include glimepiride (Amaryl), glyburide (Diabeta, Glynase PresTab, Micronase), insulin, pioglitazone (Actos), rosiglitazone (Avandia), and others.

Here's the honest truth about mixing Ceylon Leadwort with glyburide: we don't have much to go on. In animal studies, this plant has gone both ways, sometimes lowering blood sugar and sometimes raising it. That unpredictability is what makes me cautious, because glyburide already pushes your pancreas to release insulin and bring your sugar down.

So if the supplement adds to glyburide's effect, you could dip too low and feel hypoglycemia: shakiness, sweating, dizziness, confusion, or a racing heart. If it works the other way, your numbers might run higher than expected. This is only theoretical (lab and animal data, not people), so don't panic. If you want to try it, check your blood sugar closely and talk with your pharmacist or doctor first.

Ceylon Leadwort (Plumbago zeylanica) is a plant long used in Ayurvedic and other traditional medicine systems, mainly for digestion, skin problems, and pain. Solid human studies are lacking, and the plant contains plumbagin, a compound that can irritate skin and tissues and may be unsafe in larger amounts. What Is Glyburide Used For?

Brand names: Diabeta, Micronase, Glynase, Glycron, Micronized, Glynase PresTab, Albert Glyburide

Glyburide tablets are used alongside diet and exercise to help control blood sugar in adults with type 2 diabetes. It is not for type 1 diabetes or diabetic ketoacidosis (a serious complication where the body produces dangerous levels of acids called ketones).

Glyburide may be used alone or combined with metformin when metformin alone is not enough. It can also be used to help transition patients who are moving from insulin or other diabetes pills to a new regimen, under a doctor's guidance.
