Why this applies to Doxorubicin

Doxorubicin falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Ceylon Leadwort interacts with.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 3A4 (CYP3A4) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP3A4. Some drugs metabolized by CYP3A4 include lovastatin (Mevacor), clarithromycin (Biaxin), indinavir (Crixivan), sildenafil (Viagra), triazolam (Halcion), and numerous others.

Here's the idea behind this one. Doxorubicin is partly broken down by a liver enzyme called CYP3A4. A compound in Ceylon Leadwort, plumbagin, has slowed that same enzyme down in lab dish studies. If that happened in your body, doxorubicin could clear more slowly and build up, which might mean stronger effects and more side effects like mouth sores, low blood counts (raising infection risk), nausea, hair loss, and the heart strain this drug is already known for.

That said, this is only test-tube evidence and has never been seen in real people. Still, doxorubicin is serious chemotherapy with a narrow safety margin, so please don't add Ceylon Leadwort without first running it by your oncology team or pharmacist.

Doxorubicin is used to treat a wide range of cancers. It is given as part of combination chemotherapy for women with breast cancer that has spread to the lymph nodes after surgery, and it is used for many other cancers — from blood cancers like leukemia and lymphoma to solid tumors like bone and soft tissue sarcomas.

It is considered a broad-spectrum cancer fighter, meaning it is active against many different tumor types. It is almost always given alongside other chemotherapy drugs rather than on its own.
