Here is the interaction we found between these two, with the evidence behind it.
IMMUNOSUPPRESSANTS
Why this applies to Betamethasone
Betamethasone falls under IMMUNOSUPPRESSANTS — the group Melatonin interacts with.
Melatonin can stimulate immune function. Theoretically, melatonin might interfere with immunosuppressive therapy.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the thinking behind this one: betamethasone is a corticosteroid, and one of its jobs is to calm down an overactive immune response (that's how it eases the redness, swelling, and itch of conditions like eczema and psoriasis). Melatonin, on the other hand, can nudge the immune system in the opposite direction and slightly rev it up. So in theory, melatonin could work against what the steroid is trying to do.
I'll be honest with you, this is mostly a theoretical concern, and the evidence is on the weaker side. It matters far more with strong steroids taken by mouth or by injection than with a cream or ointment you rub on your skin, since very little of a topical betamethasone gets into your bloodstream. Still, it's worth a quick mention to your pharmacist or doctor, especially if you're using it over a large area or for a long time. Don't change anything on your own.

From our pharmacist’s overview on the Betamethasone page
“Betamethasone (Celestone, Diprolene) works by quieting the immune system, which is exactly why this list warrants a slow, careful read. More than fifty of the ingredients here are immune stimulants that can push back against the steroid, and another sizable group changes how much of the drug the body absorbs or holds onto.”

About Melatonin
What is Melatonin?
- Trouble falling asleep (insomnia)
- Jet lag
- Delayed sleep phase syndrome
- Non-24-hour sleep-wake disorder
- Shift-work sleep problems
- Sleep problems in some children with conditions like autism or ADHD
Melatonin is a hormone your body makes naturally to help control your sleep-wake cycle, and the supplement form is widely used to help with sleep timing problems and jet lag. The evidence is strongest for certain circadian (body-clock) disorders and is more mixed for everyday insomnia. It is generally well tolerated for short-term use, but talk with your pharmacist or doctor before starting, especially for children or during pregnancy.

Betamethasone Is Linked to Lower Levels of 8 Nutrients
Natural Medicines records Betamethasone as a medicine that can lower these. This is not an interaction with Melatonin — it is a separate effect of the medicine, and it is a reason some people are advised to supplement rather than to avoid one:
