Adrafinil is a precursor to the stimulant prescription drug modafinil. Modafinil has been shown to increase diastolic and systolic blood pressure. Retrospective analyses show that patients taking modafinil had a small but increased rate of requiring new antihypertensive drugs or increased use of antihypertensive drugs when compared with placebo.

Here's the thing to keep in mind if you take amlodipine (Norvasc) for blood pressure or chest pain. Adrafinil is converted by your liver into modafinil, a wakefulness drug that can nudge your blood pressure and heart rate upward. Amlodipine works in the opposite direction by relaxing your blood vessels to bring pressure down. So in theory, the two could work against each other, and your amlodipine might not control your numbers as well as usual.

I'll be honest, this concern is theoretical, based on how these drugs work rather than strong studies in people. Still, if you use both, it's worth checking your blood pressure at home and watching for symptoms like more frequent chest pain. Please loop in your doctor or pharmacist before combining them, and never adjust your amlodipine on your own.

Theoretically, adrafanil might reduce levels and clinical effects of CYP3A4 substrates.
Adrafinil is a precursor to the stimulant prescription drug modafinil. Modafinil has been shown to weakly induce CYP3A4 activity.

Here's the situation with these two together. Your body relies on a liver enzyme called CYP3A4 to break down amlodipine. Adrafinil turns into modafinil in your liver, and modafinil is a mild booster of that same enzyme. In theory, that could speed up how fast you clear amlodipine, which might leave a little less of it working in your system.

If that happened, your blood pressure could creep up, or your chest pain (angina) might not be as well controlled. To be fair, this is a theoretical concern based on how the drugs work, not something proven in studies, and modafinil's effect here is considered weak. Still, since adrafinil is unregulated and hard on the liver, please check with your pharmacist or doctor before combining them.

About Amlodipine

What Is Amlodipine Used For?

Brand name: Norvasc

Amlodipine treats high blood pressure (hypertension) in adults and children aged 6–17 years. Lowering blood pressure reduces the risk of strokes and heart attacks. Sdamlo (oral solution) is specifically indicated for hypertension in adults and pediatric patients 6 years of age and older.

Amlodipine also treats coronary artery disease in adults, including chronic stable angina (chest pain with exertion), vasospastic angina (Prinzmetal's or variant angina, caused by coronary artery spasms), and documented coronary artery disease in patients without heart failure or significantly reduced heart pumping function.
