Adrafinil and Aspirin, Caffeine

These two interact in 3 distinct ways. Each one is explained separately below.

Why Are There 3 Interactions?

Adrafinil interacts with whole categories of medication. Aspirin, Caffeine falls into 3 of those categories, so it can interact with Adrafinil in 3 different ways. We’ve listed each one below — your overall risk is driven by the most serious of them (Moderate).

Moderate

CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES

1 of 3

Why this applies to Aspirin, Caffeine

Aspirin, Caffeine falls under CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES — the group Adrafinil interacts with.

Theoretically, adrafinil might reduce the levels and clinical effects of CYP1A2 substrates.
Adrafinil is a precursor to the stimulant prescription drug modafinil. Modafinil has been shown to weakly induce CYP1A2 activity in vitro.
Severity Moderate Likelihood Possible Evidence D

Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.

Here's the thing about Anacin: it's mostly aspirin plus caffeine. The caffeine part is what matters here. Your liver uses an enzyme called CYP1A2 to break caffeine down. Adrafinil, once your body turns it into modafinil, may slightly speed up that enzyme. In plain terms, that could mean your body clears caffeine a little faster, so the caffeine 'kick' might feel weaker or shorter than usual.

I want to be honest that this is theoretical, based on lab studies, not real-world reports, and the effect looks weak. The aspirin portion isn't really affected. Still, since adrafinil is unregulated and rough on the liver, please run this combo by your pharmacist or doctor before mixing them.

CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES

Aspirin, Caffeine also falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES.

Theoretically, adrafanil might reduce levels and clinical effects of CYP3A4 substrates.

Dr. Brian Staiger explains PharmD, Clinical Pharmacist

Here's the gist with Adrafinil and a product like Anacin (which combines aspirin and caffeine). Once your liver turns Adrafinil into modafinil, that modafinil can mildly speed up a liver enzyme called CYP3A4. Some of the caffeine in this product is cleared partly through that pathway, so in theory your body might break it down a little faster, leaving you with slightly less of the pep you'd expect.

Honestly, this is a theoretical concern based on how these drugs work, not something proven in real patients, and the effect is described as weak. The aspirin part isn't really driven by this enzyme. Still, both Adrafinil and caffeine are stimulants, so stacking them could leave you jittery or wired. Run it by your pharmacist or doctor first.

STIMULANT DRUGS

3 of 3

Aspirin, Caffeine also falls under STIMULANT DRUGS.

Theoretically, taking adrafinil with stimulant drugs might have additive stimulant effects.
Adrafinil is a precursor to the prescription drug modafinil, which has stimulant effects.
Severity Moderate Likelihood Possible Evidence D

Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.

The product you're asking about, Anacin, combines aspirin with caffeine. The caffeine is the piece that matters here. Caffeine is a mild stimulant, and adrafinil turns into modafinil in your liver, which is also a stimulant. Stack the two together and the effects can add up.

In plain terms, you might feel more wired than you expected: a faster or pounding heartbeat, jitteriness, trouble sleeping, anxiety, or a higher blood pressure. This is a theoretical concern based on how both drugs work, not something proven in solid studies, so it doesn't mean trouble is guaranteed. Still, since adrafinil is unregulated and hard on the liver, it's worth running this by your pharmacist or doctor before combining them.

From our pharmacist’s overview on the Aspirin, Caffeine page

“Anacin combines aspirin with caffeine, so supplements can hit it from two directions, and a few of the 347 on this list deserve real respect. The biggest group, over 200 ingredients, can raise bleeding risk on top of aspirin's blood-thinning effect; beer and wine carry major ratings because alcohol plus an aspirin-type drug makes stoma…”
