Acetaminophen, Codeine, Doxylamine falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES — the group Ayahuasca interacts with.

Theoretically, ayahuasca might increase plasma concentrations of CYP2D6 substrates.
Ayahuasca contains beta-carboline alkaloids, including harmine and harmaline. In vitro research suggests that the beta-carboline alkaloids might inhibit CYP2D6 enzymes. This effect has not been reported in humans.

Here's the wrinkle with this combo, Mersyndol contains codeine, and codeine is a prodrug. It doesn't do much on its own. Your body uses an enzyme called CYP2D6 to convert it into morphine, which is what actually relieves pain. Lab studies suggest the beta-carboline compounds in ayahuasca (harmine and harmaline) can block that same enzyme. If CYP2D6 is slowed down, less codeine turns into morphine, so you may get weaker pain relief than expected.

This is theoretical and based only on test-tube data, not human studies, so I don't want to alarm you. But there's a bigger separate concern: those same compounds act as MAOIs, and mixing MAOIs with codeine or doxylamine can be genuinely dangerous. Please talk with your pharmacist or doctor before combining these.
