Chlorpheniramine, Methscopolamine, Phenylephrine falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Ceylon Leadwort interacts with.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 3A4 (CYP3A4) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP3A4. Some drugs metabolized by CYP3A4 include lovastatin (Mevacor), clarithromycin (Biaxin), indinavir (Crixivan), sildenafil (Viagra), triazolam (Halcion), and numerous others.

This combination cold and allergy product contains an antihistamine (chlorpheniramine), a drying agent (methscopolamine), and a decongestant (phenylephrine). Part of how your body clears these is through a liver enzyme called CYP3A4. In test-tube research, the plumbagin in Ceylon Leadwort appears to slow that same enzyme down. If it does the same thing inside the body, these medicines could clear more slowly and build up a bit.

In practice, that might mean stronger versions of their usual effects: more drowsiness, dry mouth, blurry vision, constipation, trouble urinating, or a faster heartbeat. But this is only theoretical, based on lab work, not real human reports, so I wouldn't lose sleep over it. Still, it's worth running past your pharmacist or doctor before combining them.

Ceylon Leadwort (Plumbago zeylanica) is a plant long used in Ayurvedic and other traditional medicine systems, mainly for digestion, skin problems, and pain. Solid human studies are lacking, and the plant contains plumbagin, a compound that can irritate skin and tissues and may be unsafe in larger amounts.
