Why this applies to Metoclopramide Hydrochloride

Metoclopramide Hydrochloride falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES — the group Ceylon Leadwort interacts with.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 2D6 (CYP2D6) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP2D6. Some drugs metabolized by CYP2D6 include amitriptyline (Elavil), codeine, desipramine (Norpramin), flecainide (Tambocor), fluoxetine (Prozac), ondansetron (Zofran), tramadol (Ultram), and others.

Your body relies on a liver enzyme called CYP2D6 to help clear metoclopramide. Ceylon Leadwort contains plumbagin, which in lab dishes slowed that same enzyme down. If that were to happen in a person, metoclopramide could build up a bit higher than expected and stick around longer.

With more metoclopramide in your system, you might notice stronger side effects like drowsiness, restlessness, or muscle twitches and stiffness. The most important thing to know is that this is only a theoretical concern from test-tube research, never actually seen in people, so I wouldn't lose sleep over it. Still, since metoclopramide already carries movement-related risks, it's worth a quick chat with your pharmacist or doctor before combining the two.

What Is Metoclopramide Nasal Spray Used For?

Brand name: Gimoti

Gimoti (metoclopramide nasal spray) is approved for adults with acute and recurrent diabetic gastroparesis — a condition where diabetes damages the nerves controlling stomach emptying, causing nausea, vomiting, and bloating. It works to relieve those symptoms by helping the stomach empty more normally.

Gimoti is not recommended for children, or for adults with moderate to severe kidney or liver disease. It is also not recommended for adults taking strong CYP2D6 inhibitor medications (certain antidepressants and other drugs that slow how the body breaks down metoclopramide).
