Olutasidenib falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Ceylon Leadwort interacts with.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 3A4 (CYP3A4) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP3A4. Some drugs metabolized by CYP3A4 include lovastatin (Mevacor), clarithromycin (Biaxin), indinavir (Crixivan), sildenafil (Viagra), triazolam (Halcion), and numerous others.

Olutasidenib (Rezlidihia) is a targeted cancer pill used for certain acute myeloid leukemias with an IDH1 mutation. Your body relies on a liver enzyme called CYP3A4 to break it down. Ceylon Leadwort contains plumbagin, which slowed down that same enzyme in lab dish studies. If that effect carried over into people, your body might clear olutasidenib more slowly, letting levels climb higher than expected.

For this drug, higher levels could mean more nausea, fatigue, liver enzyme changes, or a serious heart rhythm issue called QT prolongation. Here's the honest part: this is only test-tube evidence and has never been confirmed in humans, so don't panic. Still, with a cancer medicine like this, it's worth a quick chat with your oncologist or pharmacist before combining them.
