Valdecoxib falls under CYTOCHROME P450 2C9 (CYP2C9) SUBSTRATES — the group Ceylon Leadwort interacts with.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 2C9 (CYP2C9) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP2C9. Drugs that are metabolized by CYP2C9 include celecoxib (Celebrex), diclofenac (Voltaren), fluvastatin (Lescol), glipizide (Glucotrol), ibuprofen (Advil, Motrin), irbesartan (Avapro), losartan (Cozaar), phenytoin (Dilantin), piroxicam (Feldene), tamoxifen (Nolvadex), tolbutamide (Tolinase), torsemide (Demadex), and S-warfarin (Coumadin).

Here's the gist for Valdecoxib (Bextra). Your body relies on a liver enzyme called CYP2C9 to break this medicine down and clear it out. A compound in Ceylon Leadwort called plumbagin appears to slow that same enzyme down, at least in lab tests. If that slowdown happens in real life, Valdecoxib could build up higher than usual in your system.

More drug hanging around means a bigger chance of the side effects this type of medicine is known for, things like stomach upset, ulcers or bleeding, swelling, and higher blood pressure. I want to be honest though: this is only seen in test-tube research, not in actual people, so we're talking theory here, not a proven problem. Still, it's worth a quick chat with your pharmacist or doctor before combining the two.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 3A4 (CYP3A4) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP3A4. Some drugs metabolized by CYP3A4 include lovastatin (Mevacor), clarithromycin (Biaxin), indinavir (Crixivan), sildenafil (Viagra), triazolam (Halcion), and numerous others.

Here's the gist for Valdecoxib (Bextra). Your body partly clears this drug using a liver enzyme called CYP3A4. A compound in Ceylon Leadwort, plumbagin, has been shown in test-tube studies to slow that same enzyme down. If that happened in a real person, Valdecoxib could build up a bit higher than usual, which might mean more of its typical side effects like stomach upset, fluid retention, higher blood pressure, or increased risk of stomach irritation.

Solid human studies are lacking, and the plant contains plumbagin, a compound that can irritate skin and tissues and may be unsafe in larger amounts.
