Why this applies to Vamorolone

Vamorolone falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Ceylon Leadwort interacts with.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 3A4 (CYP3A4) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP3A4. Some drugs metabolized by CYP3A4 include lovastatin (Mevacor), clarithromycin (Biaxin), indinavir (Crixivan), sildenafil (Viagra), triazolam (Halcion), and numerous others.

Here's the situation with Vamorolone (Agamree), a steroid used mainly for Duchenne muscular dystrophy. Your body relies on an enzyme called CYP3A4 to clear it. In lab dishes, plumbagin from Ceylon Leadwort appears to slow that same enzyme down. If that were to happen in a person, Vamorolone could hang around longer and build up, which may mean stronger steroid-type effects like extra fluid retention, mood or sleep changes, higher blood sugar, and over time more bone or growth concerns.

I want to be honest though: this is only test-tube evidence, and nothing like it has been seen in real patients. So it's a theoretical caution, not a proven problem. Since Vamorolone is an important daily medicine, please check with your doctor or pharmacist before adding this supplement.
