Why this applies to Diazepam

Diazepam falls under CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES — the group Lentinan interacts with.

Laboratory evidence suggests that lentinan might increase the activity of cytochrome P450 1A2 (CYP1A2). So far, this interaction has not been reported in humans. However, watch for a decrease in the levels of drugs metabolized by CYP1A2 in patients taking lentinan.
Some drugs metabolized by CYP1A2 include amitriptyline (Elavil), haloperidol (Haldol), ondansetron (Zofran), propranolol (Inderal), theophylline (Theo-Dur, others), verapamil (Calan, Isoptin, others), and others.

Here's the gist with Valium and lentinan. Lab studies hint that lentinan may rev up a liver enzyme called CYP1A2, and diazepam is partly handled by that enzyme. If that speeds up, your body could clear the diazepam a little faster, which in theory might make it work less well. For diazepam that could mean more breakthrough anxiety, muscle tightness, or less seizure control, depending on why you take it.

That said, I want to be honest about how thin this evidence is. It comes only from test tube and animal work and has never actually been seen in people, and diazepam also leans heavily on other enzymes (CYP2C19 and CYP3A4). So this is more theoretical than proven. It's still worth a quick word with your pharmacist or doctor before adding lentinan.

Laboratory research suggests that lentinan might increase the activity of cytochrome P450 2C19 (CYP2C19). So far, this interaction has not been reported in humans. However, watch for a decrease in the levels of drugs metabolized by CYP2C19 in patients taking lentinan.
Some drugs metabolized by CYP2C19 include proton pump inhibitors including omeprazole (Prilosec), lansoprazole (Prevacid), and pantoprazole (Protonix); diazepam (Valium); carisoprodol (Soma); nelfinavir (Viracept); and others.

Lab studies hint that lentinan might rev up that enzyme, which would clear diazepam a bit faster. Diazepam works as-is (it's not a prodrug that needs activating), so faster clearance could mean it works less well, with more breakthrough anxiety, muscle spasms, or weaker seizure control.

I want to keep this in perspective, though. This is only seen in test tube and animal research, never in real patients, so it's a theoretical concern at this point. Still, if you take diazepam and are thinking about lentinan, check with your pharmacist or doctor first rather than changing anything on your own.

Laboratory research suggests that lentinan might increase the activity of cytochrome P450 3A4 (CYP3A4). So far, this interaction has not been reported in humans. However, watch for a decrease in the levels of drugs metabolized by CYP3A4 in patients taking lentinan.
Some drugs metabolized by CYP3A4 include lovastatin (Mevacor), ketoconazole (Nizoral), itraconazole (Sporanox), fexofenadine (Allegra), triazolam (Halcion), and numerous others.

If that happened in real life, your body could break diazepam down a little faster, which might leave less of it working. For some people that could mean weaker relief of anxiety, muscle spasms, or seizure control.

The important part: this has only been seen in test tubes and animals, never reported in actual patients, so it's more of a theoretical concern than something proven. Diazepam is active on its own (not a prodrug), so the direction points toward a slightly reduced effect, not a stronger one. Before adding lentinan, check with your pharmacist or doctor.

What Is Diazepam Used For?

Brand names: Valium, Valium injection, Diastat AcuDial, Valtoco

  • Diazepam is used to relieve anxiety and to control agitation caused by alcohol withdrawal. It is also used along with other medications to control muscle spasms and spasticity caused by certain neurological disorders such as cerebral palsy (condition that causes difficulty with movement and balance), paraplegia (inability to move parts of the body), athetosis (abnormal muscle contractions), and stiff-man syndrome (a rare disorder with muscle rigidity and stiffness). Diazepam is also used along with other medications to control seizures. Diazepam is in a class of medications called benzodiazepines. It works by calming abnormal overactivity in the brain.
