Here's the thing with Auvelity (the bupropion plus dextromethorphan combo): the bupropion part is well known to nudge your seizure threshold a little lower, meaning it can make seizures slightly easier to trigger in some people. This isn't about melatonin changing how your body breaks the medicine down. Instead, the concern is that melatonin itself may, in theory, raise seizure risk in certain folks, so stacking it on top of a drug that already lowers the threshold could add up.

I'll be honest, this is mostly theoretical and the strongest signal comes from children with neurological conditions, not the average adult. For most people taking Auvelity for depression, short-term melatonin for sleep is unlikely to cause trouble. Still, if you have a seizure history, please run it by your pharmacist or doctor first.

Theoretically, melatonin might increase levels of drugs metabolized by CYP2D6.
Laboratory research suggests that certain lots of melatonin inhibit CYP2D6. Theoretically, combined administration of melatonin with certain drugs metabolized by the CYP2D6 enzyme may reduce the metabolism of these drugs, resulting in increased serum levels. Until more is known, use melatonin cautiously in patients taking drugs metabolized by these enzymes.

Here's the situation with Auvelity (that's the combo of bupropion and dextromethorphan). The dextromethorphan part is normally broken down by an enzyme called CYP2D6. The bupropion part is actually there on purpose to block that enzyme, which keeps dextromethorphan levels higher so it can work in the brain. Some lab research hints that melatonin might also gently slow CYP2D6, which could theoretically nudge dextromethorphan levels up a bit more.

If that happened, you might notice things like dizziness, drowsiness, nausea, or feeling a little foggy. But honestly, this is rated minor and comes only from test-tube studies, not from people. For most folks short-term melatonin for sleep is unlikely to cause real trouble here. Still, it's worth a quick check with your pharmacist or doctor before pairing them.

Auvelity combines bupropion and dextromethorphan, and the bupropion part is actually there to boost dextromethorphan levels by blocking one of its breakdown pathways. The concern on file is that some melatonin products may lightly slow down the CYP3A4 enzyme, which also helps process these drugs. In theory, that could nudge their levels up a bit, possibly meaning more dizziness, dry mouth, nausea, jitteriness, or trouble sleeping.
