Why this applies to nilotinib

nilotinib falls under HEPATOTOXIC DRUGS — the group Ostarine interacts with.

Theoretically, concomitant use with hepatotoxic drugs might increase the risk of adverse hepatotoxic effects.
Some clinical research shows that ostarine can increase alanine aminotransferase, a marker of liver damage, in some patients. Additionally, the U.S. Food and Drug Administration warns that supplements containing SARMs, such as ostarine, have been associated with reports of liver toxicity and there are at least two reports of drug-induced liver injury attributed to the use of ostarine.

Here's the concern with pairing Ostarine and nilotinib (Tasigna): both can be hard on your liver. Nilotinib already carries a real risk of liver strain, and your oncology team checks your liver enzymes (like ALT) regularly while you take it. Ostarine, which is an unapproved SARM and not a true supplement, has also been linked to rising liver enzymes and even cases of serious liver injury.

Stacking two liver stressors together could add up. Watch for signs like yellowing of the skin or eyes, dark urine, belly pain, or unusual tiredness. To be fair, this interaction is theoretical, based on how each acts rather than studies of the two together. Still, with a serious cancer drug, I'd skip Ostarine and talk with your oncologist or pharmacist first.

nilotinib also falls under CYTOCHROME P450 3A4 (CYP3A4) INHIBITORS.

Theoretically, concomitant use of ostarine with CYP3A4 inhibitors could increase the effects and adverse effects of ostarine, although this is unlikely to be clinically significant.
Ostarine is partially metabolized by CYP3A4. However, clinical research shows that taking itraconazole, a potent inhibitor of CYP3A4, had minimal effects on the levels of ostarine in the body.

Here's the gist for these two: nilotinib (Tasigna) acts as a mild blocker of an enzyme called CYP3A4, and Ostarine is partly broken down by that same enzyme. In theory, slowing that pathway could let a little more Ostarine build up, which might mean stronger Ostarine effects and a higher chance of its known harms, like liver stress. The reassuring part is that even a powerful CYP3A4 blocker (itraconazole) barely changed Ostarine levels in studies, so this interaction is rated minor and unlikely to matter much.

My bigger concern isn't really the interaction. Ostarine is an unapproved SARM, banned in sports and tied to liver injury, and you're already on a serious leukemia drug. Please talk with your oncologist or pharmacist before mixing anything with nilotinib.

About Nilotinib

What Is Nilotinib Used For?

Brand name: Tasigna

  • Nilotinib is used to treat certain types of chronic myeloid leukemia (CML; a type of cancer of the white blood cells) who have recently found to have this condition in adults and children 1 year of age and older. It is also used to treat certain types of CML in adults whose disease could not be treated successfully with imatinib (Gleevec) or adults who cannot take imatinib because of side effects. Nilotinib is also used to treat certain types of CML in children 1 year of age or older whose disease could not be treated successfully with other tyrosine kinase inhibitor therapies or who cannot take these medications because of side effects. Nilotinib is in a class of medications called kinase inhibitors. It works by blocking the action of the abnormal protein that signals cancer cells to multiply. This helps to stop or slow the spread of cancer cells.
