Why this applies to Chlorpromazine

Chlorpromazine falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES — the group Ceylon Leadwort interacts with.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 2D6 (CYP2D6) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP2D6. Some drugs metabolized by CYP2D6 include amitriptyline (Elavil), codeine, desipramine (Norpramin), flecainide (Tambocor), fluoxetine (Prozac), ondansetron (Zofran), tramadol (Ultram), and others.

Here's the situation with these two. Chlorpromazine (Thorazine) is partly broken down by a liver enzyme called CYP2D6. Plumbagin, a compound in Ceylon Leadwort, has been shown in lab dish studies to slow that enzyme down. If that same effect happened in your body, your chlorpromazine might clear out more slowly and build up a bit higher than expected.

In real terms, that could mean stronger side effects like drowsiness, dizziness, a drop in blood pressure when you stand up, dry mouth, or muscle stiffness. I want to be honest though: this is only theoretical, based on test-tube work, and it hasn't been seen in actual people. Still, before adding this supplement, please check with your pharmacist or doctor first.

Ceylon Leadwort (Plumbago zeylanica) is a plant long used in Ayurvedic and other traditional medicine systems, mainly for digestion, skin problems, and pain. Solid human studies are lacking, and the plant contains plumbagin, a compound that can irritate skin and tissues and may be unsafe in larger amounts. What Is Chlorpromazine Used For?

Chlorpromazine is used to manage psychotic disorders and treat schizophrenia. It also controls nausea and vomiting, relieves restlessness and apprehension before surgery, and controls the manic phase of manic-depressive illness. Other uses include relief of intractable hiccups, treatment of acute intermittent porphyria, and use as an adjunct in tetanus treatment.

In children ages 1 to 12, it is used for severe behavioral problems marked by explosive or combative behavior, and for short-term treatment of hyperactive children with conduct disorders. The oral forms are also approved for the management of manifestations of psychotic disorders broadly, in addition to the uses shared with the injection form.
