Denosumab falls under IMMUNOSUPPRESSANTS — the group Ceylon Leadwort interacts with.

Plumbagin, a constituent of Ceylon leadwort, has demonstrated immunosuppressant activity in animal research. Theoretically, Ceylon leadwort might interfere with immunosuppressive therapy. Immunosuppressant drugs include azathioprine (Imuran), basiliximab (Simulect), cyclosporine (Neoral, Sandimmune), daclizumab (Zenapax), muromonab-CD3 (OKT3, Orthoclone OKT3), mycophenolate (CellCept), tacrolimus (FK506, Prograf), sirolimus (Rapamune), prednisone (Deltasone, Orasone), corticosteroids (glucocorticoids), and others.

Here's the honest picture with Denosumab (Prolia or XGEVA) and Ceylon Leadwort. Denosumab isn't broken down by liver enzymes the way most pills are. It's an injected antibody that works by quieting cells that break down bone, and because it dampens part of the immune system, it gets grouped with immunosuppressant drugs. That's the only reason this interaction shows up.

The concern is purely theoretical. In lab and animal studies, plumbagin (a compound in Ceylon Leadwort) seemed to nudge the immune system, so in theory it could tug against immune-affecting drugs like denosumab. There's no real human evidence here, so I wouldn't lose sleep over it. Still, since denosumab can raise infection risk, it's smart to mention any herbal products to your pharmacist or doctor before combining them.

Ceylon Leadwort (Plumbago zeylanica) is a plant long used in Ayurvedic and other traditional medicine systems, mainly for digestion, skin problems, and pain. Solid human studies are lacking, and the plant contains plumbagin, a compound that can irritate skin and tissues and may be unsafe in larger amounts. Talk to a pharmacist or doctor before using it, and avoid it in pregnancy.

What Is Denosumab Injection Used For?

Brand names: Prolia, XGEVA

Jubbonti (denosumab) injection is used to treat osteoporosis in postmenopausal women and men at high risk for fracture, including those who have already had a fracture or who have not responded to other osteoporosis medicines. It is also used to treat osteoporosis caused by long-term steroid (glucocorticoid) use in men and women.

Jubbonti is additionally used to increase bone mass in men receiving androgen deprivation therapy (a hormone-lowering treatment) for nonmetastatic prostate cancer, and in women receiving aromatase inhibitor therapy for breast cancer — both of which can weaken bones over time.
