Esterified Estrogens, Methyltestosterone falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Ceylon Leadwort interacts with.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 3A4 (CYP3A4) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP3A4. Some drugs metabolized by CYP3A4 include lovastatin (Mevacor), clarithromycin (Biaxin), indinavir (Crixivan), sildenafil (Viagra), triazolam (Halcion), and numerous others.

Estratest combines two hormones, esterified estrogens and methyltestosterone, and your liver uses an enzyme called CYP3A4 to help break them down. A compound in Ceylon Leadwort, called plumbagin, has slowed down this same enzyme in lab tests. If that happened in your body, these hormones could clear more slowly and build up a bit, which might mean stronger effects like breast tenderness, fluid retention, mood changes, nausea, or acne and oily skin from the testosterone part.

Here's the reassuring part: this is only seen in test-tube studies, not in real people, so it's theoretical for now. Still, since hormone levels matter, I'd run it by your pharmacist or doctor before pairing them rather than guessing.

Estratest combines an estrogen with methyltestosterone, and it's often prescribed to ease menopause symptoms like hot flashes, night sweats, and vaginal dryness. Here's the concern: in lab studies (not in people), Ceylon Leadwort root has shown anti-estrogenic activity, meaning it may push against estrogen's effects. If that played out in the body, it could theoretically blunt the benefit you're getting from the estrogen part, so your symptoms might creep back.

I want to be honest that this is only test-tube and animal evidence, so it's far from proven, and ordinary food amounts likely wouldn't matter. Still, since you're on hormone therapy, it's worth a quick chat with your pharmacist or doctor before adding a concentrated Ceylon Leadwort supplement. Don't change your Estratest on your own.
