Ethinyl Estradiol, Drospirenone falls under CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES — the group Ceylon Leadwort interacts with.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 1A2 (CYP1A2) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP1A2. Some substrates of CYP1A2 include clozapine (Clozaril), cyclobenzaprine (Flexeril), fluvoxamine (Luvox), haloperidol (Haldol), imipramine (Tofranil), mexiletine (Mexitil), olanzapine (Zyprexa), pentazocine (Talwin), propranolol (Inderal), tacrine (Cognex), zileuton (Zyflo), zolmitriptan (Zomig), and others.

Here's the short version on combining Ceylon Leadwort with Yaz (ethinyl estradiol and drospirenone). A compound in this plant called plumbagin has been shown, only in test-tube studies, to slow down an enzyme called CYP1A2. Ethinyl estradiol is partly handled by that enzyme, so in theory the hormone could build up a bit and lead to more estrogen-type side effects like nausea, breast tenderness, headaches, or bloating.

I want to be honest though: this is purely lab-based, and nothing like it has actually been seen in people. The risk here is mostly theoretical, not a known danger. Still, since solid safety data on this plant is thin, I'd run it past your pharmacist or doctor before adding it. Please don't change how you take your birth control on your own.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 3A4 (CYP3A4) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP3A4. Some drugs metabolized by CYP3A4 include lovastatin (Mevacor), clarithromycin (Biaxin), indinavir (Crixivan), sildenafil (Viagra), triazolam (Halcion), and numerous others.

Your birth control pill, Yaz, is partly cleared from your body by a liver enzyme called CYP3A4. In lab tests, plumbagin (a compound in Ceylon Leadwort) appears to slow that same enzyme down. In theory, that could let a bit more estrogen and drospirenone linger in your system, which might mean more of the usual hormone side effects like nausea, breast tenderness, bloating, or spotting.

Here is the reassuring part: this has only been seen in test tubes, not in actual people, so we really do not know if it matters in real life. Drospirenone can also nudge potassium upward, so that is one reason to be thoughtful. I would not stop either one on your own. Just give your pharmacist or doctor a heads-up before combining them.

Yaz works by delivering steady hormones (an estrogen plus a progestin) to keep you from ovulating and to keep your cycle predictable. The concern here is that lab studies hint Ceylon Leadwort root may have anti-estrogenic activity, meaning it could push against the estrogen part of your pill. In theory, that might blunt how well the hormones do their job.

I want to be honest about how thin this evidence is. This comes only from test-tube and animal research, not from real people, so we truly don't know if it matters in your body.
