Ethinyl Estradiol, Levonorgestrel falls under CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES — the group Ceylon Leadwort interacts with.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 1A2 (CYP1A2) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP1A2. Some substrates of CYP1A2 include clozapine (Clozaril), cyclobenzaprine (Flexeril), fluvoxamine (Luvox), haloperidol (Haldol), imipramine (Tofranil), mexiletine (Mexitil), olanzapine (Zyprexa), pentazocine (Talwin), propranolol (Inderal), tacrine (Cognex), zileuton (Zyflo), zolmitriptan (Zomig), and others.

Here's the gist for your birth control pill. A compound in Ceylon Leadwort called plumbagin has been shown, in test tube studies only, to slow down a liver enzyme (CYP1A2) that helps process the estrogen part of your pill. If that enzyme slows, estrogen levels could theoretically nudge upward, which can mean more of the usual estrogen-related complaints like nausea, breast tenderness, headaches, or bloating.

The important part: this has never actually been seen in real people, only in lab dishes, so the evidence is weak. Estrogen is also handled mostly by a different pathway, so the real-world effect is likely small. Still, since reliable birth control matters, I'd run it by your pharmacist or doctor before adding this supplement.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 3A4 (CYP3A4) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP3A4. Some drugs metabolized by CYP3A4 include lovastatin (Mevacor), clarithromycin (Biaxin), indinavir (Crixivan), sildenafil (Viagra), triazolam (Halcion), and numerous others.

Your birth control pill combines ethinyl estradiol and levonorgestrel, and your body relies partly on an enzyme called CYP3A4 to clear the estrogen portion. In lab dishes, plumbagin (a compound in Ceylon Leadwort) slows that enzyme down. In theory, blocking it could let estrogen levels drift a little higher than usual, which might mean more of the typical estrogen-related effects like nausea, breast tenderness, bloating, headaches, or mood changes.

I want to be honest about the strength here: this is only test-tube data, and nothing like it has actually been seen in real people taking the combo. So I would not panic.
