These two interact in 2 distinct ways. Each one is explained separately below.
Why Are There 2 Interactions?
Ceylon Leadwort interacts with whole categories of medication. Etravirine falls into 2 of those categories, so it can interact with Ceylon Leadwort in 2 different ways. We’ve listed each one below — your overall risk is driven by the most serious of them (Moderate).
CYTOCHROME P450 2C9 (CYP2C9) SUBSTRATES
1 of 2Why this applies to Etravirine
Etravirine falls under CYTOCHROME P450 2C9 (CYP2C9) SUBSTRATES — the group Ceylon Leadwort interacts with.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the short version: Ceylon Leadwort contains plumbagin, which slowed down a liver enzyme called CYP2C9 in lab dish studies. Etravirine is partly broken down by that same enzyme. So in theory, the supplement could slow how your body clears etravirine, letting its levels drift a bit higher than usual. With higher levels you might notice more of etravirine's typical side effects, such as a skin rash, nausea, or numbness and tingling.
The important part is the evidence here is weak. This was only seen in test-tube research, never confirmed in real people, so I don't want you to panic. Still, because keeping your HIV treatment steady matters a lot, please check with your pharmacist or doctor before adding Ceylon Leadwort rather than changing anything on your own.

CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES
2 of 2Etravirine also falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the gist for Etravirine (Intelence). Your body relies partly on an enzyme called CYP3A4 to break this HIV medicine down. A compound in Ceylon Leadwort called plumbagin appears to slow that same enzyme. If it does, etravirine could clear more slowly and build up a bit higher than expected, which in theory might mean more side effects like nausea, rash, or headache.
That said, I want to be honest about the evidence. This is based only on lab (test-tube) studies, and it has not actually been seen in people taking these together. So it's a possible, not proven, concern. Because keeping your HIV treatment steady matters so much, please run any Ceylon Leadwort use past your pharmacist or doctor first.

Ceylon Leadwort (Plumbago zeylanica) is a plant long used in Ayurvedic and other traditional medicine systems, mainly for digestion, skin problems, and pain. About Etravirine
What Is Etravirine Used For?
Brand name: Intelence
Etravirine (brand name Intelence) is used to treat HIV-1 infection in people who have already received other antiretroviral (HIV) treatments — meaning it is meant for patients whose HIV has had a chance to develop resistance to earlier therapies. It is always used as part of a combination regimen with other HIV medications, never on its own.
Both the brand Intelence and generic etravirine tablets are approved for adults and for children ages 2 to under 18 who weigh at least 10 kg (about 22 pounds) and who have already been treated with other HIV drugs.
