Mobocertinib Succinate falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Ceylon Leadwort interacts with.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 3A4 (CYP3A4) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP3A4. Some drugs metabolized by CYP3A4 include lovastatin (Mevacor), clarithromycin (Biaxin), indinavir (Crixivan), sildenafil (Viagra), triazolam (Halcion), and numerous others.

Here's the gist with mobocertinib (Exkivity) and Ceylon Leadwort. Your body relies on a liver enzyme called CYP3A4 to clear mobocertinib out of your system. Ceylon Leadwort's main compound, plumbagin, has been shown in lab dish studies to slow that same enzyme down. If that were to happen in real life, mobocertinib could build up higher than intended, which may worsen its known side effects, things like diarrhea, nausea, mouth sores, rash, and most importantly heart rhythm changes (a longer QT interval).

I want to be honest, though: this is theoretical. It has only been seen in test tubes, never reported in actual patients. Still, since mobocertinib is a serious cancer drug with real heart risks, I'd play it safe and check with your oncologist or pharmacist before combining them.

  • Mobocertinib is used to treat a certain type of non-small cell lung cancer (NSCLC) that cannot be removed by surgery and has spread to other parts of the body either during or after treatment with platinum chemotherapy medications. Mobocertinib is in a class of medications called kinase inhibitors. It works by blocking the action of the abnormal protein that signals cancer cells to multiply. This helps stop or slow the spread of cancer cells and may help shrink tumors.
