Why this applies to Oestrogen

Oestrogen falls under CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES — the group Ceylon Leadwort interacts with.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 1A2 (CYP1A2) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP1A2. Some substrates of CYP1A2 include clozapine (Clozaril), cyclobenzaprine (Flexeril), fluvoxamine (Luvox), haloperidol (Haldol), imipramine (Tofranil), mexiletine (Mexitil), olanzapine (Zyprexa), pentazocine (Talwin), propranolol (Inderal), tacrine (Cognex), zileuton (Zyflo), zolmitriptan (Zomig), and others.

Here's the gist: estrogen is partly broken down by a liver enzyme called CYP1A2. In lab dishes, the plumbagin in Ceylon Leadwort appears to slow that same enzyme down. If that happened in your body, less estrogen would get cleared, so the amount in your blood could creep up. With extra estrogen on board, you might notice more of its usual effects, like breast tenderness, bloating, nausea, headaches, or mood changes.

I want to be straight with you though: this is only theoretical right now, based on test-tube work, not real people. It may turn out to be a non-issue. Still, because the direction points toward higher estrogen levels, it's worth a quick chat with your pharmacist or doctor before you pair these two.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 3A4 (CYP3A4) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP3A4. Some drugs metabolized by CYP3A4 include lovastatin (Mevacor), clarithromycin (Biaxin), indinavir (Crixivan), sildenafil (Viagra), triazolam (Halcion), and numerous others.

Here's the basic idea. Your body uses a liver enzyme called CYP3A4 to help clear estrogen out of your system. A compound in Ceylon Leadwort called plumbagin has been shown to slow that enzyme down, but only in lab dish studies so far, not in actual people. If it did the same thing in your body, estrogen could be cleared more slowly and build up a bit. In theory, that might mean more of the usual estrogen-related effects, like breast tenderness, bloating, nausea, headaches, or spotting.

I want to be honest with you: this is theoretical and based on weak, early evidence. I'm not telling you to change anything. Just give your pharmacist or doctor a heads-up before combining them so they can keep an eye on you.

Here's the simple version: in lab studies, the root of Ceylon Leadwort seems to act against estrogen. Since you're taking oestrogen on purpose (for things like menopausal hot flashes, vaginal dryness, or as part of hormone therapy), a supplement pushing the opposite way could, in theory, blunt how well your treatment works. You might notice your usual symptoms creeping back.

I want to be honest about the evidence, though. This is based only on test-tube and animal research, not real people, so we truly don't know if it matters at the doses you'd take. I wouldn't lose sleep over it, but please run it past your pharmacist or doctor before combining the two so they can keep an eye on your symptoms.
