Paracetamol falls under CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES — the group Ceylon Leadwort interacts with.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 1A2 (CYP1A2) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP1A2. Some substrates of CYP1A2 include clozapine (Clozaril), cyclobenzaprine (Flexeril), fluvoxamine (Luvox), haloperidol (Haldol), imipramine (Tofranil), mexiletine (Mexitil), olanzapine (Zyprexa), pentazocine (Talwin), propranolol (Inderal), tacrine (Cognex), zileuton (Zyflo), zolmitriptan (Zomig), and others.

Here's the gist for Paracetamol (Panadol): a compound in Ceylon Leadwort called plumbagin has been shown to slow down a liver enzyme (CYP1A2) in test-tube studies. CYP1A2 is one of the pathways your body uses to process paracetamol. In theory, blocking it could let paracetamol levels run a little higher than usual.

I want to keep this in perspective. This concern comes only from lab work, not from real people, so it's theoretical at this point. Paracetamol's main safety worry is the liver when too much builds up, so if you're using both, it's worth a quick chat with your pharmacist, sticking to the recommended dose, and avoiding extra alcohol. Don't change anything on your own, just confirm it's fine for you.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 2E1 (CYP2E1) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP2E1. Some drugs metabolized by CYP2E1 include acetaminophen, chlorzoxazone (Parafon Forte), ethanol, theophylline, and anesthetics such as enflurane (Ethrane), halothane (Fluothane), isoflurane (Forane), methoxyflurane (Penthrane).

Here's the interesting twist with paracetamol (Panadol). Your liver uses an enzyme called CYP2E1 to break a small portion of it into a reactive byproduct that, in high amounts, is what actually stresses the liver. Plumbagin, a compound in Ceylon Leadwort, has been shown to block CYP2E1 in lab dishes. In theory, slowing that pathway could shift how paracetamol is processed.

But here's the honest part: this comes only from test-tube and animal work, and it has never been reported in real people. At normal, recommended paracetamol doses I wouldn't lose sleep over it. The bigger commonsense rule still stands: don't exceed the daily paracetamol limit, especially with alcohol. If you regularly use this supplement, just give your pharmacist a heads-up first.

Ceylon Leadwort (Plumbago zeylanica) is a plant long used in Ayurvedic and other traditional medicine systems, mainly for digestion, skin problems, and pain. Solid human studies are lacking, and the plant contains plumbagin, a compound that can irritate skin and tissues and may be unsafe in larger amounts. What Is Acetaminophen Injection Used For?

Brand name: Panadol

Acetaminophen Injection is given intravenously (through a vein) to manage mild to moderate pain, or moderate to severe pain when used together with opioid pain medicines, in adults and children 2 years and older. It is also used to reduce fever in adults and pediatric patients of all ages, including premature newborns born at 32 weeks gestational age or later.

For children under 2 years old, Acetaminophen Injection is approved for fever reduction, but its effectiveness for pain relief in that age group has not been established.
