Why this applies to Quizartinib

Quizartinib falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Ceylon Leadwort interacts with.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 3A4 (CYP3A4) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP3A4. Some drugs metabolized by CYP3A4 include lovastatin (Mevacor), clarithromycin (Biaxin), indinavir (Crixivan), sildenafil (Viagra), triazolam (Halcion), and numerous others.

Here's the situation with these two together. Quizartinib (Vanflyta) is a leukemia medicine that your liver clears using an enzyme called CYP3A4. In lab dishes, plumbagin from Ceylon Leadwort appears to slow that same enzyme down. If that were to happen in the body, quizartinib could build up higher than intended.

Why that matters: quizartinib already carries a warning about affecting your heart rhythm (a prolonged QT interval), and higher levels could raise the risk of that along with other side effects. But please don't panic. This is only a test-tube finding, never confirmed in people, so it's theoretical. Still, quizartinib is a serious drug with a tight safety margin, so I'd run any Ceylon Leadwort use past your oncology team or pharmacist first.
