Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 1A2 (CYP1A2) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP1A2. Some substrates of CYP1A2 include clozapine (Clozaril), cyclobenzaprine (Flexeril), fluvoxamine (Luvox), haloperidol (Haldol), imipramine (Tofranil), mexiletine (Mexitil), olanzapine (Zyprexa), pentazocine (Talwin), propranolol (Inderal), tacrine (Cognex), zileuton (Zyflo), zolmitriptan (Zomig), and others.

Here's the honest picture with Myfembree (relugolix, estradiol, and norethindrone). The concern is that plumbagin, a compound in Ceylon Leadwort, slowed down a liver enzyme called CYP1A2 in lab dishes. The estrogen part (estradiol) is partly handled by that enzyme, so in theory blocking it could let estrogen levels drift up a bit. That might mean more estrogen-type side effects like breast tenderness, nausea, headaches, bloating, or mood changes.

But I want to be clear: this is only test-tube evidence, not something seen in real patients, so I wouldn't lose sleep over it. Still, because Myfembree carries blood clot warnings, it's worth a quick chat with your pharmacist or doctor before adding this supplement.

Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP3A4. Some drugs metabolized by CYP3A4 include lovastatin (Mevacor), clarithromycin (Biaxin), indinavir (Crixivan), sildenafil (Viagra), triazolam (Halcion), and numerous others.

Myfembree combines three active medicines, and your body relies on a liver enzyme called CYP3A4 to help process them. Ceylon Leadwort contains a compound called plumbagin that, in lab studies, slowed down that same enzyme. If that effect carried over into people, the components of Myfembree could clear more slowly and build up a bit, which might mean more of the usual side effects like nausea, headache, hot flashes, breast tenderness, mood changes, or, with the estrogen and progestin parts, a theoretically higher risk of blood clots.

Here's the honest part: this is based only on test-tube research, not on real people, so we don't know if it actually happens. Still, since Myfembree already carries clotting warnings, I'd play it safe and check with your pharmacist or doctor before combining them.

Here's the thing to know with Myfembree (relugolix, estradiol, and norethindrone): one of its jobs is to deliver a steady, low dose of estrogen (estradiol) to keep your hormone levels balanced while treating fibroid or endometriosis symptoms. In lab studies only, Ceylon Leadwort root has shown anti-estrogenic activity, meaning it may push against estrogen's effects.

So in theory, taking it could blunt the estrogen part of your therapy. That might mean more hot flashes, night sweats, mood changes, or breakthrough bleeding. I'll be honest though, this is only test-tube evidence, not human studies, so we really don't know if it matters in real life. Since these hormones are carefully dosed, it's worth running it past your pharmacist or doctor before adding the supplement.

Brand name: Myfembree

This combination is used in premenopausal women. Myfembree is approved for two conditions.

It treats heavy menstrual bleeding linked to uterine fibroids (also called leiomyomas). It also treats moderate to severe pain linked to endometriosis. Use is limited to 24 months because of the risk of bone loss that may not be reversible.
