Tasimelteon falls under CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES — the group Ceylon Leadwort interacts with.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 1A2 (CYP1A2) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP1A2. Some substrates of CYP1A2 include clozapine (Clozaril), cyclobenzaprine (Flexeril), fluvoxamine (Luvox), haloperidol (Haldol), imipramine (Tofranil), mexiletine (Mexitil), olanzapine (Zyprexa), pentazocine (Talwin), propranolol (Inderal), tacrine (Cognex), zileuton (Zyflo), zolmitriptan (Zomig), and others.

Here's the gist for Hetlioz (tasimelteon): your body relies on a liver enzyme called CYP1A2 to break it down. Plumbagin, a compound in Ceylon Leadwort, has been shown in lab tests to block that same enzyme. If it does the same thing in people, your tasimelteon could clear out more slowly and build up in your system.

In practical terms, that might mean stronger effects from the medicine, like feeling more drowsy or groggy, or having a harder time waking up the next morning. The catch is that this is only theoretical right now, based on test-tube research and never reported in actual patients, so I wouldn't lose sleep over it. Still, I'd run it by your pharmacist or doctor before combining the two.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 3A4 (CYP3A4) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP3A4. Some drugs metabolized by CYP3A4 include lovastatin (Mevacor), clarithromycin (Biaxin), indinavir (Crixivan), sildenafil (Viagra), triazolam (Halcion), and numerous others.

Here's the gist with these two together. Your body clears tasimelteon using a liver enzyme called CYP3A4. Ceylon Leadwort contains a compound (plumbagin) that, in lab dishes, slowed that same enzyme down. If that happened in a real person, tasimelteon could hang around longer than usual and build up a bit.

For tasimelteon, that mostly means more of its expected effects, like extra drowsiness, grogginess the next morning, headache, or odd dreams. The important catch: this interaction has only been seen in test-tube studies, never in actual people, so it's theoretical right now. Still, since tasimelteon already affects your sleep, I'd run it by your pharmacist or doctor before pairing it with this supplement.

Ceylon Leadwort (Plumbago zeylanica) is a plant long used in Ayurvedic and other traditional medicine systems, mainly for digestion, skin problems, and pain. Solid human studies are lacking, and the plant contains plumbagin, a compound that can irritate skin and tissues and may be unsafe in larger amounts. Talk to a pharmacist or doctor before using it, and avoid it in pregnancy.

What Is Tasimelteon Used For?

Brand names: Hetlioz, Hetlioz Lq

Tasimelteon capsules are approved to treat Non-24-Hour Sleep-Wake Disorder (Non-24) in adults. Non-24 is a condition where the body's internal clock does not align with the normal 24-hour day, causing sleep and wake times to shift continuously.

Hetlioz capsules are also approved to treat nighttime sleep disturbances in Smith-Magenis Syndrome (SMS) in patients 16 years of age and older. Hetlioz LQ oral suspension is approved specifically for nighttime sleep disturbances in SMS in pediatric patients ages 3 to 15 years.
