Trazodone falls under CYTOCHROME P450 2D6 (CYP2D6) SUBSTRATES — the group Ceylon Leadwort interacts with.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 2D6 (CYP2D6) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP2D6. Some drugs metabolized by CYP2D6 include amitriptyline (Elavil), codeine, desipramine (Norpramin), flecainide (Tambocor), fluoxetine (Prozac), ondansetron (Zofran), tramadol (Ultram), and others.

Here's the gist for Trazodone: one compound in Ceylon Leadwort, called plumbagin, has slowed down a liver enzyme (CYP2D6) in test-tube studies. Your body uses that enzyme to help clear trazodone. If it gets blocked, trazodone could build up a bit more than usual, which in theory might make its effects stronger (think extra drowsiness, dizziness, dry mouth, or feeling lightheaded when you stand up).

Now, the honest part: this is only laboratory data, and it has not been seen in real people. So I wouldn't lose sleep over it, but I also wouldn't add a concentrated Ceylon Leadwort supplement without checking first. Please run it by your pharmacist or doctor before combining the two so they can look at your full med list.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 3A4 (CYP3A4) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP3A4. Some drugs metabolized by CYP3A4 include lovastatin (Mevacor), clarithromycin (Biaxin), indinavir (Crixivan), sildenafil (Viagra), triazolam (Halcion), and numerous others.

Here's the situation with these two together. Your body relies on a liver enzyme called CYP3A4 to break down and clear trazodone. A compound in Ceylon Leadwort called plumbagin has been shown to slow that same enzyme down, but only in lab dish studies so far, not in actual people.

If this effect did happen in the body, trazodone could clear more slowly and build up, which might mean stronger drowsiness, dizziness, low blood pressure when you stand up, or a groggy, hungover feeling. The good news is that the evidence here is weak and theoretical. Still, since trazodone already causes sedation, it's worth running this past your pharmacist or doctor before combining the two, especially with concentrated supplement doses.
