Why this applies to Lapatinib

Lapatinib falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Lentinan interacts with.

Laboratory research suggests that lentinan might increase the activity of cytochrome P450 3A4 (CYP3A4). So far, this interaction has not been reported in humans. However, watch for a decrease in the levels of drugs metabolized by CYP3A4 in patients taking lentinan.
Some drugs metabolized by CYP3A4 include lovastatin (Mevacor), ketoconazole (Nizoral), itraconazole (Sporanox), fexofenadine (Allegra), triazolam (Halcion), and numerous others.

Here's the gist for lapatinib (Tykerb): your liver relies on an enzyme called CYP3A4 to break this medication down. Lab and animal studies hint that lentinan may speed that enzyme up. If that held true in people, your body could clear lapatinib a little faster, which might leave less of it working against your breast cancer. The concern isn't more side effects here, it's a possible weaker cancer-fighting effect.

I want to be honest about the strength of this, though. It comes only from test-tube and animal research and has never actually been seen in humans, so it's more of a theoretical caution than a proven problem. Still, lapatinib is too important to take chances with. Please run any lentinan or shiitake supplement past your oncologist or pharmacist first.

What Is Lapatinib Used For?

Brand name: Tykerb

Lapatinib tablets — sold under the brand name Tykerb — are used to treat advanced or metastatic breast cancer in patients whose tumors overexpress the HER2 protein. They are always used in combination with another medicine, not alone.

Tykerb is approved in combination with capecitabine for HER2-positive advanced or metastatic breast cancer in patients who have already received prior treatment including an anthracycline, a taxane, and trastuzumab, and whose disease has progressed on trastuzumab. Tykerb is also approved in combination with letrozole for postmenopausal women with hormone receptor-positive, HER2-positive metastatic breast cancer when hormonal therapy is appropriate.
