These two interact in 2 distinct ways. Each one is explained separately below.
Why Are There 2 Interactions?
Lentinan interacts with whole categories of medication. Caffeine (prescription drug) falls into 2 of those categories, so it can interact with Lentinan in 2 different ways. We’ve listed each one below — your overall risk is driven by the most serious of them (Moderate).
CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES
1 of 2Why this applies to Caffeine (prescription drug)
Caffeine (prescription drug) falls under CYTOCHROME P450 1A2 (CYP1A2) SUBSTRATES — the group Lentinan interacts with.
Some drugs metabolized by CYP1A2 include amitriptyline (Elavil), haloperidol (Haldol), ondansetron (Zofran), propranolol (Inderal), theophylline (Theo-Dur, others), verapamil (Calan, Isoptin, others), and others.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the gist for caffeine. Your liver uses an enzyme called CYP1A2 to clear caffeine out of your system, and lab studies suggest lentinan may speed that enzyme up. If that happened in real life, your body might break caffeine down a bit faster, meaning it could wear off sooner and feel less effective at keeping you alert.
I want to keep this in perspective. This signal comes only from test tube and animal research, and it has never actually been reported in people. So this is more of a theoretical heads-up than a proven problem. If you notice caffeine just isn't doing much while taking lentinan, mention it to your pharmacist or doctor before changing anything on your own.

CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES
2 of 2Caffeine (prescription drug) also falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES.
Some drugs metabolized by CYP3A4 include lovastatin (Mevacor), ketoconazole (Nizoral), itraconazole (Sporanox), fexofenadine (Allegra), triazolam (Halcion), and numerous others.
Interaction has been documented in animal or in lab research, or the interaction has been documented in humans but is limited to case reports or conflicting clinical research exists.
Here's the gist for caffeine. The lab research we have suggests lentinan might speed up CYP3A4, one of the liver enzymes that helps break down certain drugs. If that held true in people, your body could clear caffeine a bit faster, meaning the alertness and pick-me-up effect might not last as long or feel as strong.
I want to be straight with you though: this is based only on test-tube and animal work, and it has never been reported in actual patients. So I wouldn't lose sleep over it (pun intended). Caffeine isn't a prodrug, so faster breakdown simply means a weaker, shorter effect, not more jitters. If you do use both regularly, just mention it to your pharmacist or doctor.

About Lentinan
What is Lentinan?
- Immune support
- Used alongside cancer treatment
- Anti-inflammatory
- General wellness/tonic
- Antimicrobial support
Lentinan is a beta-glucan polysaccharide from shiitake mushrooms that is studied mainly as an immune-modulating add-on to cancer chemotherapy, where it is given by injection in a few countries. Evidence for oral supplements is limited, and quality varies widely between products. Talk with your pharmacist or doctor before using it, especially if you have a health condition or are pregnant or breastfeeding.
About Caffeine
What Is Caffeine Used For?
Brand names: Dextrophin, Caffeine
Caffeine citrate injection and oral solution are used for the short-term treatment of apnea of prematurity. This means pauses in breathing in premature infants. The label lists the age range as infants between 28 weeks and a later gestational age, and that range is cut off in the text provided.
Over-the-counter caffeine tablets, such as Diurex and the Stay Awake products, are taken by mouth by adults and older children. Their labels point to fatigue and drowsiness. Diurex directions also mention taking it while symptoms persist.
