Why this applies to Everolimus

Everolimus falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Lentinan interacts with.

Laboratory research suggests that lentinan might increase the activity of cytochrome P450 3A4 (CYP3A4). So far, this interaction has not been reported in humans. However, watch for a decrease in the levels of drugs metabolized by CYP3A4 in patients taking lentinan.
Some drugs metabolized by CYP3A4 include lovastatin (Mevacor), ketoconazole (Nizoral), itraconazole (Sporanox), fexofenadine (Allegra), triazolam (Halcion), and numerous others.

Here's the gist for everolimus. Your body relies on a liver enzyme called CYP3A4 to break it down and clear it out. Lab studies hint that lentinan may speed that enzyme up. If that happened in people, your everolimus could be cleared a little faster, which might leave less drug in your system and could mean it doesn't work as well against the cancer or tumors it's treating.

I want to be honest about the evidence here: this comes only from test tube and animal research, and it has not been seen in actual patients. So it's a theoretical concern, not a proven one. Still, everolimus is a drug where blood levels really matter, so loop in your oncologist or pharmacist before adding lentinan rather than starting it on your own.

What Is Everolimus Used For?

Brand names: Certican, Afinitor, Zortress, Afinitor Disperz

Everolimus is used to treat several serious conditions. In cancer, it is prescribed for advanced hormone receptor-positive, HER2-negative breast cancer (in postmenopausal women), certain neuroendocrine tumors of the pancreas, GI tract, or lung, and advanced renal cell carcinoma (kidney cancer) when other treatments have stopped working.

It is also used for people with tuberous sclerosis complex (TSC) — a genetic condition that causes benign tumors to grow throughout the body. In TSC, it treats kidney tumors called angiomyolipomas, brain tumors called SEGA, and seizures. In kidney transplant patients, it helps prevent the body from rejecting the new organ.
