Why this applies to Modafinil

Modafinil falls under CYTOCHROME P450 3A4 (CYP3A4) SUBSTRATES — the group Lentinan interacts with.

Laboratory research suggests that lentinan might increase the activity of cytochrome P450 3A4 (CYP3A4). So far, this interaction has not been reported in humans. However, watch for a decrease in the levels of drugs metabolized by CYP3A4 in patients taking lentinan.
Some drugs metabolized by CYP3A4 include lovastatin (Mevacor), ketoconazole (Nizoral), itraconazole (Sporanox), fexofenadine (Allegra), triazolam (Halcion), and numerous others.

Here's the gist for Modafinil. Your body uses a liver enzyme called CYP3A4 to help clear modafinil out. Lab and animal studies hint that lentinan might rev up that enzyme. If that held true in people, your body could break modafinil down a little faster, which might soften its wake-promoting punch. In real life, that could look like more daytime sleepiness creeping back in or the medicine just not feeling as effective as usual.

I want to be honest about the strength here: this is only test-tube and animal data, and it has not been seen in actual patients. So I wouldn't lose sleep over it (pun intended). Still, if you start lentinan and notice your modafinil isn't working like it should, give your pharmacist or doctor a heads-up before changing anything.

What Is Modafinil Used For?

Brand names: Provigil, Sparlon, Alertec

Modafinil (also sold as Provigil) is used to improve wakefulness in adults who experience excessive sleepiness due to narcolepsy, obstructive sleep apnea (OSA), or shift work disorder (SWD).

In OSA, modafinil treats the sleepiness — not the underlying airway obstruction itself. It is used alongside standard treatments such as CPAP (a breathing device worn during sleep), not as a replacement for them. If CPAP is appropriate for you, your doctor will want you to use it consistently even while taking modafinil.
