Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 1A2 (CYP1A2) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP1A2. Some substrates of CYP1A2 include clozapine (Clozaril), cyclobenzaprine (Flexeril), fluvoxamine (Luvox), haloperidol (Haldol), imipramine (Tofranil), mexiletine (Mexitil), olanzapine (Zyprexa), pentazocine (Talwin), propranolol (Inderal), tacrine (Cognex), zileuton (Zyflo), zolmitriptan (Zomig), and others.

Here's the simple version. Your body relies mostly on a liver enzyme called CYP1A2 to clear caffeine out of your system. Ceylon Leadwort contains plumbagin, which in lab dishes slowed that same enzyme down. If that effect were to happen in a person, caffeine could hang around longer and pile up, giving you stronger jittery effects like a racing or pounding heart, restlessness, trouble sleeping, an upset stomach, or feeling wired and anxious.

The honest part: this is only seen in test-tube research, never confirmed in real people, so it's theoretical. I wouldn't lose sleep over it, but if you're using concentrated Ceylon Leadwort supplements, run it past your pharmacist or doctor first to be safe.

Plumbagin, a constituent of Ceylon leadwort, has been shown to inhibit cytochrome P450 3A4 (CYP3A4) in vitro. So far, this interaction has not been reported in humans. Theoretically, Ceylon leadwort might increase the levels of drugs metabolized by CYP3A4. Some drugs metabolized by CYP3A4 include lovastatin (Mevacor), clarithromycin (Biaxin), indinavir (Crixivan), sildenafil (Viagra), triazolam (Halcion), and numerous others.

Here's the thing to keep in mind with this pair. Ceylon Leadwort contains plumbagin, which in lab dishes slowed down a liver enzyme called CYP3A4. The injectable caffeine and sodium benzoate product is mostly cleared by a different enzyme (CYP1A2), so plumbagin's effect on CYP3A4 is likely a minor player here at best.

That said, if caffeine levels did creep up, you might feel the usual caffeine overshoot: a racing or pounding heart, jitteriness, trouble sleeping, or an upset stomach. The honest truth is that this interaction is only theoretical, based on test-tube work and never reported in people. Still, it's worth a quick chat with your pharmacist or doctor before combining the two, especially since this caffeine form is usually given in a hospital setting.
